產(chǎn)品介紹:
TRIpure 試劑是直接從細胞或組織中提取總RNA的試劑。它在破碎和溶解細胞時能保持RNA的完整性。加入氯仿后離心,樣品分成水樣層和有機層。RNA存在于水樣層中。收集上面的的水樣層后,RNA可以通過異丙醇沉淀來還原。在除去水樣層后,樣品中的DNA和蛋白也能相繼以沉淀的方式還原。乙醇沉淀能析出中間層的DNA,在有機層中加入異丙醇能沉淀出蛋白。共純化DNA對于樣品間標(biāo)準(zhǔn)化RNA的產(chǎn)量十分有用。
無論是人、動物、植物還是細菌組織,該方法對少量的組織(50-100mg)和細胞(5×106)以及大量的組織(≥1g)和細胞(>107)均有較好的分離效果。TRIpure 試劑操作上的簡單性允許同時處理多個的樣品。所有的操作可以在一小時內(nèi)完成。TRIpure抽提的總RNA能夠避免DNA和蛋白的污染。故而能夠作RNA印跡分析、斑點雜交、poly(A)+選擇、體外翻譯、RNA酶保護分析和分子克隆。如果是用于PCR,當(dāng)兩條引物位于單一外顯子內(nèi)時,建議用擴增級的DNase I來處理抽提的總RNA。
TRIpure 試劑能促進不同種屬不同分子量大小的多種RNA的析出。例如,從大鼠肝臟抽提的RNA瓊脂糖凝膠電泳并用溴化乙啶染色,可見許多介于7 kb和15 kb之間不連續(xù)的高分子量條帶(mRNA和hnRNA成分),兩條優(yōu)勢核糖體~5 kb (28S)和~2 kb(18S),低分子量RNA介于0.1和0.3 kb之間 (tRNA, 5S)。當(dāng)抽提的RNA用TE稀釋時其A260/A280比值≥1.8。
